I have a client who is a Trustee of a charity. The client wishes to donate some of his own money into the charity. Is there anything stopping him from making a Gift Aid claim on his own tax return? Does the fact that he is a Trustee of that charity block a claim?

yes

My understanding is that they can but that the transactions have to actually be made.

For example very often Trustees do not claim travel or similar expenses. But if such expenses were claimed and paid and disclosed as necessary in the accounts and the trustee made a donation of the same amout to the charity then gift aid could be claimed by both te charity and the taxpayer.
